I recommend that you unlock most upgrades and weapons before taking on the ArachnoGauntlets. Have at least cargo shorts equipped, as well as the tracker upgrades SPIDAR and Arachnoscope to have an easier time tracking down spiders

First Contact:
Use your energy drinks to make the most of your time. Crouch down for a better chance to hit the spiders.
Close Encounter:
Energy Drinks help here as well.
Domestic Duties:
Spritz objects and walls with the flamethrower or hairspray
Down The Spider Hole:
The shoe may be underpowered when you first get it, just keep replaying the gauntlet until the shoe gains explosive powers.
Major Inconvenience:
Detect and suck spiders and debris into the vaccuum prior to taking on the challenge, then toss them out in one spot.
Garden of Evil:
Use the energy drinks, and make sure to dodge the web shots! Giant Magnet upgrade reccommended
Barnstormer:
Pop an energy drink and make sure you have the flashlight on
Paper Trails:
With Cargo shorts, you should have enough room for 8 weapons. Know where the spiders spawn to kill them faster
Contingency Omega:
Slap the ground the ground and panel with an upgraded shoe
Consequences:
With Cargo pants, cover EVERYTHING in gasoline. Once you start the challenge, light the gasoline and keep pouring! Shoe strat also works.
